目的观察寒凝血瘀模型大鼠下丘脑神经递质的变化及其对生殖内分泌的调节和影响。方法将大鼠置于0~1℃冰水中造成妇科寒凝血瘀证大鼠模型,采用放射免疫分析方法测定血清孕酮(P)、雌二醇(E2)、睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)的含量及下丘脑β-内啡肽(β-EP)含量,采用荧光分光光度法测定下丘脑5-羟色胺(5-HT)、去甲肾上腺素(NA)、多巴胺(DA)含量。结果模型组大鼠的血清E2、P、T、LH、FSH明显降低,下丘脑β-EP、5-HT、NA、DA的含量明显升高,与正常组比较有显著性差异(P<0.01或P<0.05)。结论下丘脑分泌的神经介质β-EP、NA、DA、5-HT参与了生殖内分泌的调节和寒凝血瘀证的形成。
Objective To observe the change of hypothalamus neurotransmitter and its regulation and effect on reproductive and endocrine in rats with cold and blood stasis syndrome. Methods Rats were placed in 0 ~ 1 ℃ ice water to induce gynecological cold coagulation and blood stasis in rats. Radioimmunoassay was used to determine the serum progesterone (P), estradiol (E2), testosterone (T), follicle stimulating hormone (FSH), luteinizing hormone (LH), and beta-endorphin (β-EP) in the hypothalamus were measured by spectrofluorometry. The levels of 5-hydroxytryptamine (5-HT), norepinephrine ), Dopamine (DA) content. Results The levels of E2, P, T, LH and FSH in the model group were significantly decreased, and the contents of β-EP, 5-HT, NA and DA in the hypothalamus were significantly higher than those in the normal group Or P <0.05). Conclusion The neurotransmitters β-EP, NA, DA and 5-HT secreted by the hypothalamus are involved in the regulation of reproductive and endocrine processes and the formation of cold-coagulation and blood stasis syndrome.
