为了比较不同家系凡纳滨对虾应对环境胁迫差异,试验采用逆境胁迫法及酶性测定方法分析了7个家系凡纳滨对虾M3期对低氧和氨氮的耐受力差异,同时测定了对低氧耐受性差异显著的两个家系(6203与正大)不同组织(鳃、肝胰腺、肌肉与血淋巴)中免疫酶活性的时空变化特征。结果表明:各家系对低氧胁迫12 h的半数致死浓度(LC50)变化范围为2.22~2.97 mg/L,对氨氮24 h的LC50变化范围为34.18~39.39 mg/L,对应的非离子氨LC50变化范围为1.59~1.83 mg/L。酚氧化物酶(PO)含量变化趋势为鳃>肌肉>血>肝胰腺,过氧化物酶(POD)活性变化趋势为血>鳃>肝胰腺>肌肉,总超氧化物歧化酶(T-SOD)活性变化趋势为鳃>血>肌肉>肝胰腺。免疫相关酶活性具有组织与家系特异性,鳃组织中免疫相关酶响应最敏感,可以作为应答低氧耐受性的重要指标。
In order to compare the differences of response to environmental stress among different families of Litopenaeus vannamei, the tolerance to hypoxia and ammonia nitrogen in seven families was studied using stress tolerance and enzymatic determination methods. Spatiotemporal characteristics of the immune enzyme activity in different tissues (gill, hepatopancreas, muscle and hemolymph) of two families (6203 vs. Chia Tai) with significant differences in oxygen tolerance were observed. The results showed that the LC50 of each line at 12 h of hypoxia stress ranged from 2.22 to 2.97 mg / L, the LC50 of ammonia-nitrogen at 24 h ranged from 34.18 to 39.39 mg / L, and the corresponding non-ionic ammonia The LC50 ranged from 1.59 to 1.83 mg / L. Immune-related enzyme activity is tissue-specific and family-specific, and immune-related enzymes in gill tissue respond most sensitively and may serve as an important indicator of hypoxia tolerance.
