目的本实验采用免疫组织化学方法研究了Survivin在乳腺癌组织及癌前病变中的表达情况,并探讨其与突变型P53蛋白表达之间的关系。方法采用免疫组织化学ABC法检测了10例正常乳腺组织,42例乳腺癌组织(其中浸润性导管癌31例,髓样癌6例,腺癌4例,派杰氏病1例)和乳腺增生组织34例(包括普通增生11例,不典型增生23例)中Survivin、P53蛋白表达的情况,探讨其与突变型P53蛋白表达之间的关系,以揭示Survivin在乳腺癌发生、发展过程中的作用。结果Survivin在乳腺癌组织及癌前病变中表达的组织化学记分和阳性率显著高于正常乳腺组和普通增生组(P<0.01),P53蛋白在乳腺癌及不典型增生组的阳性率显著高于正常组和普通增生组(P<0.05);P53蛋白表达阳性组Survivin的组织化学记显著高于P53蛋白表达阴性组(P<0.05)。结论Survivin蛋白的异常表达与乳腺癌的发生与发展密切相关,Survivin蛋白表达与P53基因突变相关,两者可能通过某种作用共同参与乳腺癌的发生与发展。
Objective To investigate the expression of Survivin in breast cancer tissues and precancerous lesions using immunohistochemistry and to explore its relationship with the expression of mutant P53 protein. Methods Immunohistochemical ABC method was used to detect the expression of 10 normal breast tissues, 42 breast cancer tissues (including 31 invasive ductal carcinomas, 6 medullary carcinomas, 4 adenocarcinoma and 1 Papillary’s disease) and breast hyperplasia To investigate the expression of Survivin and P53 in 34 cases (including 11 cases of common hyperplasia and 23 cases of dysplasia), and to explore the relationship between the expression of Survivin and P53 protein in order to reveal the occurrence and development of Survivin in breast cancer effect. Results The positive rate of Survivin expression in breast cancer tissues and precancerous lesions was significantly higher than that in normal breast tissues and normal proliferative tissues (P <0.01). The positive rate of P53 protein in breast cancer and atypical hyperplasia was significantly higher (P <0.05). The histochemistry of Survivin in P53 protein positive group was significantly higher than that in P53 negative group (P <0.05). Conclusion The abnormal expression of Survivin protein is closely related to the occurrence and development of breast cancer. The expression of Survivin protein is related to the mutation of P53 gene, both of which may play an important role in the occurrence and development of breast cancer.
