目的 :检测纤维粘连蛋白 (FN)及其受体整合素 β1 亚单位在子宫内膜癌组织中的表达 ,探讨与组织分级和临床分期的关系以及在子宫内膜癌转移浸润中的意义。方法 :用免疫组织化学技术和图像分析法检测 5 8例子宫内膜腺癌、8例正常内膜组织中 FN和 β1 整合素的表达。结果 :正常子宫内膜腺上皮细胞 FN弱阳性表达 ,主要分布于细胞基底部 ;内膜腺癌组织 FN染色增强 ,明显高于正常对照组 (P<0 .0 1) ;低分化腺癌染色强度低于高分化组 (P<0 .0 5 ) ,阳性面积比例与细胞分级成反比。 β1 整合素在正常子宫内膜腺上皮呈强阳性表达 ;在不同分化的内膜腺癌组织表达均明显减少 (P<0 .0 1) ;与细胞分化有关 ;FN和 β1 整合素的表达与临床分期无关。结论 :子宫内膜腺癌组织 FN和 β1 整合素表达与正常明显不同。抑制肿瘤细胞的转移应采用调节粘附蛋白和粘附分子表达的策略。
OBJECTIVE: To detect the expression of fibronectin (FN) and its receptor integrin β1 subunit in endometrial carcinoma and to investigate its relationship with tissue grading and clinical stage and its significance in metastasis and invasion of endometrial carcinoma. Methods: Immunohistochemistry and image analysis were used to detect the expression of FN and β1 integrins in 58 cases of endometrial adenocarcinoma and 8 cases of normal endometrium. Results: The expression of FN in normal endometrial glandular epithelial cells was weakly positive and mainly distributed in the base of cells. The FN staining of endometrial adenocarcinoma was significantly higher than that in normal control group (P <0.01). The staining of poorly differentiated adenocarcinoma The intensity was lower than that of the well-differentiated group (P <0.05), and the positive area ratio was inversely proportional to the cell grade. β1 integrin in normal endometrial glandular epithelium was strongly positive expression in different differentiated endometrial adenocarcinoma tissue were significantly decreased (P <0.01); and cell differentiation; FN and β1 integrin expression and Clinical stage has nothing to do. Conclusion: The expression of FN and β1 integrin in endometrial adenocarcinoma is obviously different from normal. Inhibition of tumor cell metastasis should be used to regulate the adhesion protein and adhesion molecule expression strategy.
