作者证明,硝苯吡啶吸入对人类组织胺引起的支气管收缩具有轻度的保护作用,但不改变休息时气道的张力。由于体外试验硝苯吡啶对气道平滑肌收缩前的松弛作用比对收缩的保护作用更有效,所以研究了吸入硝苯吡啶对组织胺引起收缩的恢复作用。6例轻度哮喘患者(几何均数PC_(35)=2.3mg/ml的组织胺),年龄20~38岁,通过剂量测定计给予一个剂量的组织胺,以降低特异性比气道传导率(sGaw),用装有电子计算机的体积描记器,按大约以50%进行测定。然后采用随意双盲法,3天时间内分别给予40%乙醇中含硝苯呲啶5mg、舒喘灵2.5mg,或单独吸入40%乙醇等的气雾剂。测定sGaw在吸入后立即和60分钟时进行。
The authors showed that inhaled nifedipine had mild protective effects on bronchoconstriction induced by histamine in humans but did not alter the airway tension at rest. Since nifedipine in vitro is more effective than contractile protection in relaxing the airway smooth muscle contraction, the effect of inhaled nifedipine on the recovery of histamine induced contractions was studied. Six patients with mild asthma (histamine with geometric mean PC 35 = 2.3 mg / ml), aged 20-38 years, were given a dose of histamine via a dosimeter to reduce the specific specific airway conductance (sGaw), using a computerized plethysmograph, at approximately 50%. Then use the random double-blind method, three days were given within 40% ethanol containing nifedipine 5mg, salbutamol 2.5mg, or inhalation of 40% ethanol alone aerosol. SGaw was determined immediately after inhalation and 60 minutes.
