目的:筛选出最佳的有机溶剂,使血液中的盐酸川芎嗪得到最大的富集,并进一步优选最佳的萃取条件。方法:采用高效液相色谱法测定,研究4种不同极性的有机溶剂(乙醇、三氯甲烷、二氯甲烷、石油醚)对血液中盐酸川芎嗪的萃取后,血液中盐酸川芎嗪的浓度。结果:最佳的富集溶剂是三氯甲烷,最佳萃取条件是:溶剂量7 ml,超声时间12 min,涡旋时间3 min。结论:采用三氯甲烷优选条件下萃取能够使血液中的盐酸川芎嗪得到最大的富集,为进一步研究新型付罐促渗法对盐酸川芎嗪凝胶剂中盐酸川芎嗪透皮吸收家兔体内的血药浓度变化提供定量方法。
Objective: To screen out the best organic solvent to maximize the enrichment of tetramethylpyrazine hydrochloride in the blood and to optimize the extraction conditions. Methods: After the extraction of ligustrazine hydrochloride in blood by four different polar organic solvents (ethanol, chloroform, methylene chloride and petroleum ether), the concentration of ligustrazine in the blood . Results: The best solvent for enrichment was chloroform. The optimal extraction conditions were as follows: solvent 7 ml, sonication time 12 min, vortexing time 3 min. CONCLUSION: The extraction of ligustrazine hydrochloride in blood by chloroform extraction is the best. To further study the effect of the new method of promoting canister infusion on ligustrazine hydrochloride penetration in rabbits by ligustrazine hydrochloride The blood concentration changes provide a quantitative method.
