本文利用流式细胞仪研究了雷公藤总甙对人T细胞亚群及对IL-2产生和IL-2RTac链表达的影响。结果证实T_Ⅱ可明显抑制PHA刺激的T细胞IL-2RTac链表达,抑制IL-2产生,且均有剂量依赖关系。T_Ⅱ对T细胞亚群中CD细胞和CD细胞从G_0/G_1期到S+G_2/M期均有明显的抑制作用。但两者受抑制的程度不同,其中对CDT细胞的抑制作用更强。提示雷公藤总甙治疗自身免疫病的机制之一可能与其抑制IL-2自泌环路,降低体内活化的T细胞数量,调整T细胞亚群比例,从而恢复异常的免疫状态有关。
In this paper, flow cytometry was used to study the effects of Tripterygium wilfordii on human T cell subsets and the production of IL-2 and the expression of IL-2RTac chain. The results confirmed that T_II can significantly inhibit PHA-stimulated T cell IL-2RTac chain expression and inhibit IL-2 production in a dose-dependent manner. T_II significantly inhibited CD and CD cells in T cell subsets from the G 0 /G 1 phase to the S + G 2 /M phase. However, both are inhibited to different extents, among which the inhibitory effect on CDT cells is stronger. It is suggested that one of the mechanisms of treatment of autoimmune diseases by Tripterygium wilfordii may be related to its inhibition of IL-2 autocrine loop, lowering the number of activated T cells in vivo, adjusting the ratio of T cell subsets, and thus restoring abnormal immune status.
