研究胶原蛋白-壳聚糖-海藻酸盐(Collagen-chitosan-alginate,CCA)复合敷料的促愈合效果,并初步评价生物安全性。制备大鼠背部全皮层创伤模型,以医用纱布为阴性对照,市售壳聚糖敷料为阳性对照,术后不同时期取材并进行组织学和EGF、b FGF、TGF-β和CD31免疫组化染色观察。采用CCK-8细胞毒性实验进行初步安全性评价。CCA复合敷料能促进创面愈合,术后d 7,CCA组伤口愈合率为(70.26±1.39)%,显著高于纱布组的(60.59±0.71)%(P<0.05),极显著高于壳聚糖组的(58.38±0.68)%(P<0.01)。术后d 12,CCA组愈合率达到(98.32±0.48)%。免疫组化结果表明,术后d 5,CCA组的EGF、b FGF、TGF-β表达率达到峰值,提前于纱布组。CD31染色结果表明,CCA组新生血管生成于术后d 5最活跃,极显著高于纱布组(P<0.01)。各浓度的敷料浸提液细胞增殖率均大于75%,毒性等级为一级,无细胞毒性。CCA复合敷料具有显著的促愈合作用,有望开发为高效医用敷料。
To study the healing effect of collagen-chitosan-alginate (CCA) composite dressing and to evaluate the bio-safety. The rat model of full-thickness cortical injury was established. The medical gauze was used as the negative control. The chitosan dressing was used as the positive control. The histological and EGF, b FGF, TGF-β and CD31 immunohistochemical staining Observed. CCK-8 cytotoxicity experiments were used to evaluate the initial safety. The wound healing rate of CCA group was (70.26 ± 1.39)%, significantly higher than that of gauze group (60.59 ± 0.71)% (P <0.05), which was significantly higher than that of chitosan (58.38 ± 0.68)% of the sugar group (P <0.01). At 12 days after operation, the healing rate in CCA group was (98.32 ± 0.48)%. The results of immunohistochemistry showed that the expression of EGF, b FGF and TGF-β in CCA group reached the peak at d 5, which was earlier than that in gauze group. The results of CD31 staining showed that the angiogenesis in CCA group was the most active at d5, significantly higher than that in gauze group (P <0.01). All concentrations of dressing extract cell proliferation rate of more than 75%, a toxic level, no cytotoxicity. CCA composite dressing has a significant role in promoting healing and is expected to be developed as a highly effective medical dressing.
