目的 观察分析弓形虫表面抗原 P2 2编码基因真核表达质粒 (p BK/P2 2 )接种小鼠诱导的细胞免疫效果。方法 采用肌肉注射免疫接种 BAL B/c小鼠 ,5周后 ,取鼠脾细胞作 Con A刺激淋转试验 (MTT法 )及 T细胞亚群的测定。结果 MTT试验中 ,p BK/P2 2免疫组、空质粒 p BK- CMV对照组与 NS对照组的 Con A孔的光密度值与不加 Con A孔的光密度值的差值疫苗免疫组略高于二对照组 ,统计学分析无明显差异 (P>0 .0 5 ) ;CD+4 细胞数量变化无显著性差异 (P>0 .0 5 ) ,而 CD+8细胞数量则明显增多 ,p BK/P2 2免疫组、空质粒 p BK- CMV均高于 NS对照组 ,其差异有显著性 (P<0 .0 1) ;p BK/P2 2免疫组与空质粒组间无显著性差异 (P>0 .0 5 )。结论 p BK/P2 2对 Con A刺激的淋巴细胞增殖功能作用不明显 ;T淋巴细胞亚群测定结果表明 ,p BK/P2 2免疫后小鼠 CD+4 淋巴细胞增殖不明显 ,而 CD+8淋巴细胞则显著增殖
Objective To observe and analyze the cellular immunity induced by inoculation of pBK / P2 2 into mice induced by P2 2 gene of Toxoplasma gondii surface antigen. Methods BALB / c mice were immunized with intramuscular injection. After 5 weeks, splenocytes were taken for Con A stimulation test (MTT) and T cell subsets. Results In the MTT assay, the differences between the optical density values of Con A wells and those without Con A wells of pBK / P2 2 immunized group, empty plasmid p BK-CMV control group and NS control group (P> 0.05). There was no significant difference in the number of CD + 4 cells between the two groups (P> 0.05), but the number of CD + 8 cells was significantly increased pBK / P2 2 immunized group, empty plasmid pBK-CMV were higher than the NS control group, the difference was significant (P <0.01); pBK / P2 2 immunohistochemistry and empty plasmid group was not significant Differences (P> 0.05). Conclusion The effect of p BK / P2 2 on the proliferation of lymphocytes stimulated by Con A was not obvious. The results of T lymphocyte subsets showed that the proliferation of CD 4 + lymphocytes was not obvious after immunization with p BK / Lymphocytes significantly proliferate
