目的:观察雷公藤提取物对大鼠心脏急性损伤的影响。方法:将SD大鼠随机分为空白对照组和雷公藤提取物低(137 mg/kg)、中(275 mg/kg)、高(550 mg/kg)剂量组,各组ig给药,分别于给药前(0 h)和给药后1、2、4、8、12、24h检测血钾、心电图及心功能指标,并采用HE染色法观察24h大鼠心肌组织病理变化。结果:与给药前或空白对照组比较,各剂量组均出现血钾降低、心电图指标:心率降低,Q-T间期、S-T间期、QRS间期延长和ST段下降;心功能指标:左室收缩压(LVSP)降低、左室内压最大上升/下降速度(±dp/dtmax)降低、左室舒张末压(LVEDP)升高。各指标均在4~8h变化最显著,且给药剂量越大,各指标变化出现时间越早,变化程度越明显。HE染色显示,给药后低剂量组大鼠心肌未见病理变化,中剂量组大鼠心肌有轻微损伤、高剂量组大鼠心肌有明显损伤。结论:雷公藤提取物在137~550 mg/kg剂量范围内灌胃给药后对大鼠心脏有急性损伤作用,且给药剂量越大,损伤越明显。血钾降低可能为心脏急性损伤的原因之一。
Objective: To observe the effects of Tripterygium extraction on acute heart injury in rats. Methods: SD rats were randomly divided into blank control group and Tripterygium extraction (137 mg / kg), medium (275 mg / kg) and high (550 mg / kg) Blood potassium, electrocardiogram and cardiac function were measured before administration (0 h) and at 1, 2, 4, 8, 12 and 24 hours after administration. Pathological changes of myocardium were observed by HE staining. Results: Compared with the pre-administration group or the blank control group, the serum potassium levels were decreased in all dose groups. The ECG indexes such as heart rate decreased, QT interval, ST interval, QRS interval and ST segment decreased. LVSP, ± dp / dtmax, LVEDP increased. The indexes all changed most significantly in 4 ~ 8h, and the larger the dose, the earlier the changes of each index occurred, the more obvious the degree of change. HE staining showed that there was no pathological changes in the myocardium of the low dose group after administration, the myocardial damage in the middle dose group was mild, and the myocardial damage in the high dose group was obvious. CONCLUSION: Tripterygium wilfordii extract has an acute injury on rat heart after intragastric administration of 137. 550 mg / kg dose, and the greater the dose, the more obvious the injury. Reduced serum potassium may be one of the causes of acute heart injury.
