为探讨硒对甲醛致A549细胞激活蛋白-1(AP-1)和核转录因子-κB(NF-κB)表达的影响,以A549细胞株为实验对象,设对照组、0.1 mmol/L甲醛组、1.25 mg/L硒+0.1 mmol/L甲醛组、2.5 mg/L硒+0.1 mmol/L甲醛组、5.0 mg/L硒+0.1 mmol/L甲醛组、10.0 mg/L硒+0.1 mmol/L甲醛组分别进行染毒,采用蛋白免疫印记法检测核蛋白中AP-1、NF-κB的表达。结果显示,对照组A549细胞生长活跃,1.25、2.5、5.0、10.0 mg/L硒+0.1 mmol/L甲醛组染毒12、24、48 h后细胞生长受到抑制,且抑制率随硒浓度和染毒时间的增加而升高;0.1 mmol/L甲醛可使A549细胞核蛋白中AP-1和NF-κB的表达增加,而2.5、5.0、10.0 mg/L硒+0.1 mmol/L甲醛组可抑制AP-1和NF-κB的过度表达,与单纯甲醛染毒组比较,差异有统计学意义(P<0.05)。提示硒在一定浓度下能够拮抗甲醛致A549细胞AP-1和NF-κB的表达。
In order to investigate the effect of selenium on the expression of AP-1 and NF-κB in A549 cells, the A549 cell line was used as the experimental group. The control group, the 0.1 mmol / L formaldehyde group , 1.25 mg / L selenium + 0.1 mmol / L formaldehyde, 2.5 mg / L selenium + 0.1 mmol / L formaldehyde, 5.0 mg / L selenium + 0.1 mmol / L formaldehyde, 10.0 mg / L selenium + 0.1 mmol / L Formaldehyde group were exposed to the virus, Western blot analysis of AP-1, NF-κB expression. The results showed that the A549 cells in the control group grew actively, and the cell growth was inhibited at 1.25, 2.5, 5.0 and 10.0 mg / L selenium + 0.1 mmol / L for 12, 24 and 48 h, 0.1 mmol / L formaldehyde could increase the expression of AP-1 and NF-κB in A549 cell nucleus, while the inhibitory effect of AP in APF-treated A549 cells treated with 2.5, 5.0, 10.0 mg / L Selenium + 0.1 mmol / L formaldehyde -1 and NF-κB overexpression, compared with formaldehyde-only group, the difference was statistically significant (P <0.05). It suggested that selenium could antagonize the expression of AP-1 and NF-κB in A549 cells induced by formaldehyde.
