目的 研究磷脂酰肌醇3激酶的特异性抑制剂Wortmannin( WT) 对细胞的放射增敏效应,以探索放射增敏信号传导途径。方法 采用体外细胞克隆培养法观察不同浓度WT 在不同时间对3 种细胞的放射增敏效应。结果 50 μmol/L以上浓度的WT 在照射后6 小时内对LP3 细胞呈较明显的增敏效应,剂量存活曲线D0 增敏比在2 左右;50 μmol/L WT 对HeLa 细胞和SP/20细胞均呈明确增敏效应。结论 WT是一种新型放射增敏剂。
Objective To study the radiosensitizing effect of Wortmannin (WT), a specific inhibitor of phosphatidylinositol 3 kinase, on radiosensitization signal transduction pathway. Methods The effects of different concentrations of WT on the proliferation of three kinds of cells were observed by cell clone culture in vitro. Results WT at a concentration above 50 μmol / L showed a significant sensitizing effect on LP3 cells within 6 hours after irradiation, and the D0 sensitization ratio of the dose-survival curve was about 2. In 50 μmol / L WT cells, the proliferation of HeLa cells and SP / 20 cells Clear sensitization were showed. Conclusion WT is a new radiosensitizer.
