为了更好的保护并促进优异葡萄种质资源的有效区分和合理利用,利用SSR标记技术对酿酒葡萄品种(系)材料中选取的15份特异性资源进行亲缘关系的分析、分类、种质识别。从44对SSR引物中筛选出15对用于供试葡萄种质的SSR扩增,共扩增出74条带,其中多态性条带71条,多态性百分率为95.9%。根据SSR扩增结果,分析表明15份葡萄材料遗传距离的变异范围为0~1.092 4。UPGMA聚类分析表明,在遗传相似系数0.63处,可将15份供试材料分为2个类群,准确地检测出基因型之间的遗传背景与遗传关系。通过多态性谱带的有序编码转换,构建了15个葡萄品种的分子身份证系统,结果表明,利用SSR标记进行酿酒葡萄种质资源的鉴定和保护是可行的。
In order to better protect and promote the effective differentiation and rational utilization of excellent grape germplasm resources, SSR markers were used to analyze and classify 15 specific germplasm resources of selected grapevine varieties (strains) . Fifteen out of 44 SSR primers were screened for SSR amplification of grape germplasms tested. A total of 74 bands were amplified, of which 71 were polymorphic bands with a percentage of 95.9%. According to the results of SSR amplification, the analysis showed that the variation range of genetic distance of 15 grape materials was 0 ~ 1.092 4. UPGMA cluster analysis showed that at the genetic similarity coefficient of 0.63, 15 test materials could be divided into two groups to accurately detect the genetic background and genetic relationship between genotypes. The molecular ID card system of 15 grape cultivars was constructed through the sequential coding transformation of polymorphic bands. The results showed that it is feasible to identify and protect the grape germplasm resources by SSR markers.
