以不同花色、不同发育时期的鸳鸯茉莉(Brunfelsia acuminate Benth.)花瓣为材料,通过单因素试验方法,分别以不同浓度及pH值的Tris-HCl和磷酸缓冲液(PBS),获得提取花瓣中过氧化物酶(POD)的最佳浓度和pH值,并对不同花色、不同发育时期花瓣的POD活性进行测定。结果分别选出0.4 mol/L Tris-HCl,pH7.0和0.1 mol/L PBS,pH7.0 2种缓冲液组合,其中以后者的提取效果最好,并通过POD同工酶谱得以证明;不同发育时期POD活性:白花>淡紫花>紫花,花瓣颜色褪变与POD活性呈正相关,且差异均达显著水平(p<0.05);花瓣中不同形态的POD活性之间差异显著,可溶态POD活性远大于结合态的POD活性。
Using petals of Brunfelsia acuminate Benth. From different colors and different developmental stages, the petals of Brunfelsia acuminate Benth. Petal were extracted by different concentrations and pH values of Tris-HCl and phosphate buffered saline (PBS) by single factor test The optimum concentration and pH value of POD were determined, and the POD activity of petals with different color and different developmental stages was determined. Results Two kinds of buffer solutions, 0.4 mol / L Tris-HCl, pH7.0 and 0.1 mol / L PBS, pH7.0, were selected respectively. Among them, the latter had the best extraction effect and was proved by POD isozymes. There was a positive correlation between POD activity and POD activity at different developmental stages (P> 0.05). There were significant differences in POD activity between different developmental stages POD activity is much greater than that of bound POD activity.
