目的:探讨麝香酮抗痫作用及可能的分子生物学机制。方法:将健康雄性SD大鼠90只随机分成6组:空白对照组、模型组、麝香酮小剂量组、麝香酮中剂量组、麝香酮大剂量组及丙戊酸钠组,每组各15只。通过行为学、免疫组化、Western Blot及q RT-PCR方法观察大鼠癫痫发作及脑内c-jun表达情况,以及不同剂量麝香酮对其干预作用。结果:空白对照组大鼠无癫痫发作,脑组织中仅有少量c-jun表达,而模型组及麝香酮小剂量组大鼠癫痫发作级别较高,脑皮质及海马中有大量c-jun表达。麝香酮中剂量组及大剂量组大鼠癫痫发作级别较模型组及小剂量组低(P<0.05,P<0.01),c-jun表达减少(P<0.01)。与麝香酮中剂量组比较,麝香酮大剂量组大鼠癫痫发作级别更低(P<0.05,P<0.01),c-jun表达减少更加明显(P<0.01)。结论:麝香酮具有一定的抗痫作用,且呈量效关系,其机理可能与阻断脑内c-jun表达有关。
Objective: To investigate the anti-epileptic effects of musk ketone and possible molecular biological mechanisms. Methods: 90 healthy male SD rats were randomly divided into 6 groups: control group, model group, low-dose musk ketone group, middle musk ketone group, high-dose musk ketone group and sodium valproate group, with 15 only. The behavioral, immunohistochemical, Western Blot and q RT-PCR methods were used to observe the expression of c-jun and epileptic seizures in rats, and the effect of different doses of musk ketone on it. Results: There was no seizure in the blank control group, and only a small amount of c-jun was expressed in the brain tissue. In the model group and the low-dose muskone group, the level of seizure was higher in rats with c-jun expression in cortex and hippocampus . The levels of epileptic seizures in musk ketone middle-dose group and high-dose group were lower than those in model group and low-dose group (P <0.05, P <0.01), while the expression of c-jun was decreased (P <0.01). Compared with the muskone medium dose group, rats in the high-dose musk ketone group had lower levels of seizure (P <0.05, P <0.01) and decreased c-jun expression (P <0.01). Conclusion: Musk ketone has a certain anti-epileptic effect, and the dose-response relationship, the mechanism may be related to block c-jun expression in the brain.
