目的建立同时测定独活中蛇床子素和二氢欧山芹醇当归酸酯含量的反相高效液相色谱法。方法 Shiseido C18MG(4.6 mm×250 mm,5μm)色谱柱,流动相乙腈-水(49∶51),检测波长330 nm,流速1.0 mL.min-1,柱温30℃。结果蛇床子素在0.008 2~0.656 mg.mL-1内呈良好线性,r=1.000 0,平均回收率99.6%(RSD=2%);二氢欧山芹醇当归酸酯在0.003 6~1.536 mg.mL-1内呈良好线性,r=0.999 9,平均回收率99.4%(RSD=2.2%)。结论通过对22批不同来源独活药材与饮片的含量测定,证明该方法简单,准确可靠,重复性好。在重庆巫山地区以海拔1 600~1 700 m、向阳、黑土的生长条件下,独活中蛇床子素和二氢欧山芹醇当归酸酯的含量均较高。
OBJECTIVE To establish an RP-HPLC method for the simultaneous determination of Osthol and Dihydropalmatine angelica in indoxan. Methods The mobile phase consisted of acetonitrile - water (49:51) with a detection wavelength of 330 nm and a flow rate of 1.0 mL · min-1 on a Shiseido C18MG (4.6 mm × 250 mm, 5 μm) column at 30 ℃. Results Osthol showed a good linearity in the range of 0.008 2-0.656 mg · mL-1, r = 1.000 0 with an average recovery of 99.6% (RSD = 2%). Paeoniflorin was 0.003 6-1.536 mg. mL-1, r = 0.999 9, with an average recovery of 99.4% (RSD = 2.2%). Conclusion The determination of 22 batches of independent living herbs and Pieces, proved that the method is simple, accurate and reliable, reproducible. In the Wushan area of Chongqing, the contents of Osthol and Dihydropalmatine angelate were higher under the conditions of sunny and black soil growth at the elevation of 1 600 ~ 1 700 m.
