目的观察角膜内皮细胞(CEC)对淋巴细胞活化和增殖的抑制活性,并分析其机制。方法通过植片技术分离大鼠和兔CEC。在CEC单层上,观察淋巴细胞对丝裂原刀豆蛋白A(ConA)和同种异体抗原刺激的增殖反应。另外,将CEC培养上清直接转移到由ConA刺激的淋巴细胞增殖系统以观察CEC释放某些因子的抑制效应。结果CEC对ConA或同种异体抗原刺激的淋巴细胞增殖反应可以施加有效的抑制效应。在这些检测系统中所观察到的抑制效应不具有MHC限制性。将CEC培养上清直接转移到由ConA刺激的淋巴细胞增殖系统,并未观察到显著的抑制效应。然而,当CEC培养上清经过浓缩和酸处理后,则在胸腺细胞共刺激实验中显示剂量-反应形式的抑制效应。结论CEC通过细胞交互作用和(或)释放某些抑制性因子对淋巴细胞的活化和增殖发挥潜在的抑制作用。CEC可能是构成眼前房免疫赦免的因素之一。
Objective To observe the inhibitory activity of corneal endothelial cells (CECs) on the activation and proliferation of lymphocytes and to analyze its mechanism. Methods Rat and rabbit CECs were isolated by the suture technique. On CEC monolayers, the proliferative response of lymphocytes to mitogen-induced concanavalin A (ConA) and alloantigen stimulation was observed. In addition, CEC culture supernatants were directly transferred to lymphocyte proliferation system stimulated by ConA to observe the inhibitory effect of certain factors released by CEC. Results CEC could exert a potent inhibitory effect on the proliferation of lymphocytes stimulated by ConA or alloantigen. The inhibitory effects observed in these detection systems are not MHC-limited. The CEC culture supernatant was directly transferred to the lymphocyte proliferation system stimulated by ConA, and no significant inhibitory effect was observed. However, when the CEC culture supernatant was concentrated and acid-treated, the inhibitory effect of the dose-response form was shown in thymocyte costimulation experiments. Conclusion CEC plays a potential role in the activation and proliferation of lymphocytes through cell-cell interaction and / or release of certain inhibitory factors. CEC may be one of the factors that constitute animmune immunotherapy in anterior chamber.
