目的:通过检测脾气虚证模型大鼠回肠组织中线粒体呼吸链酶复合物Ⅰ、Ⅳ活性,MDA含量,GSH-Px活性的变化,探讨脾气虚证大鼠自由基代谢和细胞能量代谢的调控机制。方法:选取SPF级大鼠39只,随机方式分为健康对照组、脾气虚证模型组和脾气虚证中药反证组,每组均13只。采用经典复合因素塑造脾气虚证大鼠模型。行比色法检测每组大鼠回肠组织中丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-Px)活性以及回肠组织线粒体呼吸链复合物Ⅰ、Ⅳ活性。结果:(1)脾气虚模型组MDA含量明显高于正常组(P<0.05),GSH-Px,线粒体呼吸链酶复合物Ⅰ、Ⅳ活性明显低于正常组(P<0.05)。(2)脾气虚中药反证组MDA含量低于脾气虚证模型组(P<0.05),GSH-Px,线粒体呼吸链酶复合物Ⅰ、Ⅳ活性活性高于模型组(P<0.05)。结论:在脾气虚证状态下大鼠,自由基代谢和细胞能量代谢出现异常,GSH-Px,线粒体呼吸链酶复合物Ⅰ、Ⅳ的活性降低,脂质过氧化物MDA的含量增高,补中益气汤能有效干预以上机制,提示补中益气汤可能对自由基修复和细胞能量代谢障碍存在保护机制。
OBJECTIVE: To investigate the changes of mitochondrial respiratory chain complex Ⅰ, Ⅳ, MDA content and GSH-Px activity in the rat ileum of spleen qi deficiency syndrome model and to explore the regulation mechanism of free radical metabolism and cellular energy metabolism in rats with spleen-qi deficiency syndrome . Methods: Thirty-nine SPF rats were randomly divided into three groups: healthy control group, spleen-qi deficiency syndrome model group and spleen-qi deficiency syndrome syndrome control group, with 13 in each group. Using classical compound factors to establish the rat model of spleen deficiency syndrome. The contents of malondialdehyde (MDA), glutathione peroxidase (GSH-Px), and mitochondrial respiratory chain complex I and IV in ileum of rats in each group were determined by colorimetry. Results: (1) The content of MDA in spleen deficiency model group was significantly higher than that in normal group (P <0.05). The activities of GSH-Px and mitochondrial respiratory chain complex Ⅰ and Ⅳ were significantly lower than those in normal group (P <0.05). (2) The content of malondialdehyde (MDA) in TCM syndrome of spleen deficiency syndrome was lower than that of the model group of spleen qi deficiency syndrome (P <0.05). The activities of GSH-Px and mitochondrial respiratory chain complex Ⅰ, Ⅳ of mitochondrial enzymes were higher than those of model group (P <0.05). CONCLUSION: The metabolism of free radicals and cell energy in rats with spleen-qi deficiency syndrome is abnormal. The activities of GSH-Px and mitochondrial respiratory chain enzyme complexes Ⅰ and Ⅳ are decreased and the content of lipid peroxides MDA is increased. Yiqi Decoction can effectively interfere with the above mechanism, suggesting that Bu Zhong Yi Qi Decoction may have a protective mechanism for free radical repair and cellular energy metabolism disorders.
