建立了比格犬血浆样品中托烷司琼的反相高效液相色谱检测法。血浆样品 ,加入盐酸三环哌酯作内标 ,用乙酸乙酯提取 ,经稀盐酸反洗 ,再用二氯甲烷提取。色谱条件 :C8柱 ;流动相 :甲醇 -水 (5 5∶4 5 ) ,加 0 3%三乙胺 ,用冰醋酸调pH 7 2 ;检测波长 2 83nm。线性范围 5 4 5~70 0ng/mL ,最低检测浓度为 3 0ng/mL ,回收率为 79 9%~ 85 8% ,日内RSD为 1 0 6 %~4 2 7% ,日间RSD为 2 94 %~ 9 89%。采用本法对比格犬口服托烷司琼的药代动力学进行了研究。
A reverse phase HPLC method was established for tropisetron in beagle dog plasma samples. Plasma samples were added with tricyclic hydrochloride as internal standard, extracted with ethyl acetate, back-washed with dilute hydrochloric acid and extracted with dichloromethane. Chromatographic conditions: C8 column; mobile phase: methanol - water (5 5:45), add 0 3% triethylamine, adjust pH 7 2 with glacial acetic acid; The linear range was 545 ~ 70 0 ng / mL, the lowest detection concentration was 30ng / mL, the recovery was 79 9% ~ 85 8%, the intraday RSD was 106% ~ 427%, and the intraday RSD was 2 94 % ~ 9 89%. The pharmacokinetics of oral tropisetron in beagle dogs were studied by this method.
