为了建立一种新的培养嗅鞘细胞的方法,从而为神经诱导修复材料的研究提供种子细胞。本研究取新生鼠的嗅球最外两层经胰蛋白酶消化成单细胞悬液,经差速贴壁法纯化,观察并记录其形态特征;经HE染色以及神经生长因子蛋白受体p75(NGFR-p75)和S-100免疫组化染色鉴定并计算其纯度。结果显示,获得的嗅鞘细胞突起呈双极或三级,p75和S-100阳性细胞纯度达到91%。上述结果提示该方法经济易行,所获得的嗅鞘细胞纯度高、活性好。
In order to establish a new method of culturing olfactory ensheathing cells, seed cells were provided for the study of nerve-induced repair materials. In this study, the outermost two layers of the olfactory bulb of newborn rats were trypsinized into single cell suspension and purified by differential adherence. Morphological characteristics were observed and recorded. HE staining and NGFR- p75) and S-100 immunohistochemistry to identify and calculate the purity. The results showed that the olfactory ensheathing cells were bipolar or tertiary, and the purity of p75 and S-100 positive cells was 91%. The above results suggest that the method is economical and feasible, and the obtained olfactory ensheathing cells have high purity and good activity.
