目的研究JNK在Na As O2诱导L-02肝细胞凋亡中的作用。方法用不同浓度Na As O2染毒L-02肝细胞,采用流式细胞术检测L-02肝细胞凋亡率;Western-blot检测L-02肝细胞中Jnk、p-Jnk、Caspase-3蛋白的相对表达量。结果 0、50、100和150μmol/L组细胞凋亡率分别为3.33%±0.30%、7.49%±0.23%、9.48%±0.49%和14.87%±2.17%,染毒组的细胞凋亡率随Na As O2浓度的增加而升高,差异有统计学意义(P<0.05);染毒组细胞内Jnk、p-Jnk、Caspase-3蛋白相对表达量均增高,差异有统计学意义(P<0.05)。结论 Na As O2能诱导L-02肝细胞凋亡的发生,这可能与激活JNK,增加JNK磷酸化水平,继而激活凋亡调控基因Caspase-3有关。
Objective To investigate the role of JNK in Na As O2-induced L-02 hepatocyte apoptosis. Methods L-02 hepatocytes were exposed to different concentrations of Na As O2 and the apoptotic rates of L-02 hepatocytes were detected by flow cytometry. Jnk, p-Jnk and Caspase-3 proteins were detected by Western-blot in L-02 hepatocytes Relative expression level. Results The apoptotic rates of 0, 50, 100 and 150 μmol / L groups were 3.33% ± 0.30%, 7.49% ± 0.23%, 9.48% ± 0.49% and 14.87% ± 2.17% (P <0.05). The relative expression of Jnk, p-Jnk and Caspase-3 protein in the cells increased significantly, the difference was statistically significant (P < 0.05). Conclusions Na As O2 can induce the apoptosis of L-02 hepatocytes, which may be related to the activation of JNK, the increase of JNK phosphorylation, and the activation of apoptosis-regulating gene Caspase-3.
