在培养基中加入一定浓度的Nacl,对水稻F_2代成熟胚愈伤组织的耐盐突变体进行细胞水平上的筛选,采用逐渐加盐浓度多次继代筛选法。结果表明:此方法是提高耐盐变异体频率的有效措施。诱导愈伤组织Na十Nacl0.3%继代3次的愈伤组织→Ms分化频率达15.74%,获得了在0.3%含盐土壤上种植能正常生长的耐盐株系四个。
A certain concentration of Nacl was added to the culture medium to screen the salt-tolerant mutants of the callus from the mature embryos of F 2 generation in rice. The results show that this method is an effective measure to increase the frequency of salt-tolerant mutants. The callus induction rate of Ms + 10% Na + 0.3% was 15.74% after 3 times of subculture, and four salt-tolerant lines with normal growth on 0.3% salt-tolerant soil were obtained.
