目的:观察小半夏加茯苓颗粒含药血清对肝癌SMMC-7721细胞凋亡及凋亡相关基因Bcl-2表达的影响。方法:选用SD大鼠制备小半夏加茯苓颗粒及原方汤剂、环磷酰胺(CTX)含药血清。①采用ELISA法检测细胞凋亡相关基因Bcl-2表达。②FCM法观察细胞生长凋亡情况。结果:①与空白对照组比较,颗粒剂组细胞液中Bcl-2含量降低(P<0.05),汤剂组显著降低(P<0.01)。②与同时段内空白对照组比较,含药血清作用18 h后,环磷酰胺(CTX)组、汤剂组、颗粒剂组细胞存活率有所下降(P<0.05);作用36 h后,CTX组、汤剂组细胞存活率显著下降(P<0.01)。结论:小半夏加茯苓颗粒含药血清可下调肝癌SMMC-7721细胞Bcl-2基因表达,抑制其生长增殖,其作用机制可能与诱导细胞凋亡或细胞毒作用有关。
Objective: To observe the effect of Xiaobanxia plus Fuling granule serum on the apoptosis of hepatocellular carcinoma cell line SMMC-7721 and the expression of apoptosis related gene Bcl-2. Methods: SD rats were prepared small Banxiaxia Fuling granules and the original Decoction, cyclophosphamide (CTX) containing serum. ①Apoptosis-related gene Bcl-2 expression was detected by ELISA. ② FCM observation of cell growth and apoptosis. Results: ①Compared with the blank control group, the Bcl-2 level in the granule group decreased significantly (P <0.05), and decoction group decreased significantly (P <0.01). ②Compared with blank control group in the same time period, the cell viability of CTX group, decoction group and granule group decreased after 18 h of treatment (P <0.05); after 36 h of treatment, CTX group, decoction group cell viability decreased significantly (P <0.01). Conclusion: Xiaobanxia plus Fuling granule containing serum can down-regulate the expression of Bcl-2 gene in hepatocellular carcinoma SMMC-7721 cells and inhibit its growth and proliferation, the mechanism may be related to the induction of apoptosis or cytotoxicity.
