目的 建立一种快速、灵敏和可靠的气相色谱法以评价氟康唑原位胶化眼部给药系统兔眼应用后氟康唑的生物利用度。方法 应用大口径毛细管柱。检测为氮磷检测器。将泪液处理后和房水用甲醇溶解注入色谱仪中。采用外标法定量。结果 每个样品的分析时间小于10min,最小可检测浓度为0.01μg/ml,在0.1至20.0μg/ml范围内线性关系良好。药物从泪液和房水中的回收率分别为99.0%和94.8%。结论 此方法可用于测定兔眼泪液和房水中的氟康唑浓度,对于评价氟康唑局部应用后眼部生物利用度是一个可行的、快速的、有效可靠的方法。
Objective To establish a rapid, sensitive and reliable gas chromatography for the evaluation of the bioavailability of fluconazole after the application of fluconazole in situ ophthalmic ophthalmic drug delivery system in rabbits. Results The analysis time of each sample was less than 10 min, the minimum detectable concentration was 0.01 μg / ml, and the linearity was good in the range of 0.1 to 20.0 μg / ml. Drug recovery from tear fluid and aqueous humor were 99.0% and 94.8%, respectively. Conclusion This method can be used to determine the concentration of fluconazole in rabbit tears and aqueous humor. It is a feasible, rapid, effective and reliable method for the evaluation of ocular bioavailability after topical application of fluconazole.
