以‘津研四号’黄瓜品种为材料,克隆得到一个黄瓜膜联蛋白基因的cDNA全长,命名为CsANN2。为进一步探究黄瓜CsANN2基因的功能,对CsANN2基因编码的蛋白序列进行生物信息学分析后,发现其理化性质、保守结构域以及多个功能蛋白位点都和其他植物的膜联蛋白一致。系统进化树分析表明,黄瓜CsANN2与AtANN2、AtANN6、AtANN7、BjANN2和BjANN6有较近的亲缘关系。亚细胞定位结果显示,CsANN2蛋白在细胞膜、细胞质与细胞核内都有分布。qRT-PCR结果分析表明,CsANN2基因受ABA、SA、PEG和H2O2诱导表达,但对NaCl胁迫无响应。
Using the ’Jinyan No.4’ cucumber variety as material, a full-length cucumber annexin gene was cloned and named as CsANN2. In order to further explore the function of CsANN2 gene in cucumber, bioinformatics analysis of the protein sequence encoded by CsANN2 gene revealed that its physical and chemical properties, conserved domains and multiple functional protein sites were consistent with those of other plant annexin. Phylogenetic tree analysis showed that cucumber CsANN2 had close genetic relationship with AtANN2, AtANN6, AtANN7, BjANN2 and BjANN6. Subcellular localization results showed that CsANN2 protein was distributed in the cell membrane, cytoplasm and nucleus. Analysis of qRT-PCR results showed that CsANN2 gene was induced by ABA, SA, PEG and H2O2, but did not respond to NaCl stress.
