[目的]比较大肠菌群荧光定量PCR方法和多管发酵法检测结果的相关性。[方法]采用建立的大肠菌群荧光定量PCR方法和多管发酵法同时检测系列浓度梯度的实验室模拟水样的大肠菌群含量。对结果进行相关性分析,比较两种方法定量结果的相关性。[结果]大肠菌群荧光定量PCR能够能准确检测高于102cfu/100ml水平的水质大肠菌群含量,在此水平以上的定量结果同多管发酵法结果具有较高相关性。[结论]大肠菌群荧光定量PCR方法具有快速、特异、操作简便的特点,可以作为多管发酵法检测方法的补充,用于水质大肠菌群的快速定量检测。
[Objective] To compare the correlation between coliforms fluorescence quantitative PCR method and multi-tube fermentation method. [Method] The concentration of coliform bacteria in laboratory simulated water samples of series of concentration gradients was simultaneously detected by the established coliforms fluorescence quantitative PCR method and multi-tube fermentation method. Correlation analysis of the results, the correlation between the two methods quantitative results. [Result] Coliform bacteria fluorescence quantitative PCR could accurately detect the water quality coliform bacteria above 102cfu / 100ml level, and the quantitative results above this level had a higher correlation with the results of multi-tube fermentation. [Conclusion] The colony fluorescence quantitative PCR method is rapid, specific and easy to operate. It can be used as a supplement to the multi-tube fermentation method for the rapid and quantitative detection of coliform bacteria in water quality.
