滨蒿内酯对哮喘豚鼠血清和肺组织匀浆中白细胞介素4与干扰素γ失衡的调节作用(英文)

背景:滨蒿内酯是传统中药茵陈的主要有效成分之一,具有免疫抑制、消炎和舒张支气管平滑肌的作用。滨蒿内酯是否能改善哮喘豚鼠血清及肺组织匀浆中白细胞介素4与干扰素γ(IL-4/IFN-γ)失衡、进而抑制IgE的产生而发挥平喘作用目前尚不清楚。目的:研究滨蒿内酯的平喘作用及其对哮喘豚鼠免疫功能的调节作用。设计:以实验动物为研究对象,随机对照的验证性研究。单位:一所大学的天然药物研究室。材料:实验于2000-07/2002-03在中国医科大学药理教研室完成。选择健康雄性豚鼠84只,体质量200±50)g,清洁级。(干预:分别采用10g/L组胺与1g/L乙酰胆碱混合液引喘及10mL/L卵蛋白致敏复制法制备急性和迟发性哮喘豚鼠模型,采用雾化吸入途径给予滨蒿内酯药治疗,观察引喘潜伏期,并分别采用化学发光法、放免法和双抗夹心法检测迟发性哮喘豚鼠血清及肺组织匀浆中IgE,IL-4和IFN-γ的含量。主要观察指标:引喘潜伏期以及实验动物血清及肺组织匀浆中IgE,IL-4和IFN-γ含量。结果:滨蒿内酯明显延长急性和迟发性哮喘引喘潜伏期,哮喘组豚鼠血清及肺组织匀浆中IgE,IL-4含量较对照组明显增加(P<0.05),IFN-γ含量明显降低(P<0.05);而滨蒿内酯组IgE水平及IL-4含量较哮喘组豚鼠显著降低(P<0.05),IFN-γ的含量则明显升高(P<0.05)。结论:滨? BACKGROUND: The scoparone lactone is one of the main active ingredients of the traditional Chinese medicine capillaris, and has the effects of immunosuppression, anti-inflammatory and diastolic bronchial smooth muscle. Whether scoparone lactone can improve the imbalance of interleukin-4 and interleukin-4 (IL-4/IFN-γ) in serum and lung tissue homogenate of asthmatic guinea pigs and suppress the production of IgE and exert antiasthmatic effect is still unclear. Objective: To study the antiasthmatic effect of scoparone lactone and its regulation of immune function in asthmatic guinea pigs. Design: A randomized, controlled, validated study of experimental animals. Unit: A university’s natural medicine laboratory. MATERIALS: The experiment was performed at the Department of Pharmacology, China Medical University from July 2000 to March 2002. 84 healthy male guinea pigs with a body mass of 200±50 g were selected and cleaned. (Intervention: Acute and delayed asthmatic guinea pig models were prepared using 10 g/L histamine and 1 g/L acetylcholine mixture as inducers and 10 mL/L egg protein sensitized replication method, respectively. After treatment, the latency of asthma was observed, and the levels of IgE, IL-4 and IFN-γ in the serum and lung homogenate of the late-onset asthmatic guinea pigs were detected by chemiluminescence, radioimmunoassay, and double-antibody sandwich assay, respectively.
