用15%脂肪中饱和脂肪酸占78、68、48、38%的半合成饲料喂饲大鼠,测定血浆及肝中脂质的合量,吸收率,淋巴中脂蛋白的脂肪酸组成,各种脂蛋白成份及脱辅基蛋白,以电镜观察淋巴脂蛋白的形状与体积,用X线衍射、核磁共振光谱仪及差示扫描热量计测定脂蛋白的物理性能。 其结果表明:大鼠血浆中HDL胆固醇偏高,有较强的胆固醇廓清作用,所以大鼠不是临床高脂血症理想的动物模型。膳食中饱和脂肪酸百分比降低时,脂肪颗粒变大,吸收率增加,颗粒体积与吸收率成显著相关(r=0.99)。控制脂蛋白代谢的脱辅基蛋白在饱和脂肪酸百分比降低时Ar明显增加,C降低即LCAT酶活力增加。说明膳食中饱和脂肪酸较低时,胆固醇转移酶活跃,脂蛋白在周围组织中即可被廓清。物理性能的测定表明,膳食中饱和脂肪酸比例在68%以上时,机体淋巴是以相对稳定的,过度冷却的液相存在。因而过去常规方法在4—20℃条件下提取与分离脂蛋白是不恰当的。
Rats were fed with 78,68,48,38% of semi-synthetic feed containing saturated fatty acids in 15% fat. The amount of lipids in plasma and liver, absorption rate, fatty acid composition of lymphatic lipoproteins, Protein composition and apoprotein were observed by electron microscope. The shape and volume of lymphatic lipoprotein were observed by electron microscope. The physical properties of lipoprotein were determined by X-ray diffraction, nuclear magnetic resonance spectroscopy and differential scanning calorimetry. The results showed that the rat plasma HDL cholesterol is high, there is a strong role of cholesterol clearance, so the rat is not an ideal animal model of hyperlipidemia. When the percentage of saturated fatty acids in the diet decreased, the fat particles became larger and the absorption rate increased. The volume of the particles was significantly correlated with the absorption rate (r = 0.99). Arginine, which controls the metabolism of lipoproteins, significantly increases Ar when the percentage of saturated fatty acids decreases, while C decreases, indicating an increase in LCAT enzyme activity. Description dietary saturated fatty acid is low, cholesterol transfer enzyme is active, lipoproteins in the surrounding tissue can be cleared. Measurement of physical properties showed that when the proportion of saturated fatty acids in the diet is above 68%, the lymphatic tissues are present in a relatively stable and overcooled liquid phase. Therefore, in the past conventional methods at 4-20 ℃ extraction and separation of lipoprotein is not appropriate.
