目的:观察鼻咽癌(NPC)患者外周血单核细胞来源的树突状细胞(DC)的形态、表型和功能与健康者DC的差别。方法:从12例NPC患者(NPC组)和9例健康者(对照组)的外周血中分离和培养DC,观察DC的形态,用流式细胞仪检测DC的表型CD83、CD1a、CD86和HLA-DR,用4,二甲基噻唑2,5二苯基四唑溴化物(MTT)比色法检测DC诱导混合淋巴细胞反应(MLR)的能力。结果:对照组的DC形态较NPC组成熟,NPC组CD83、CD1a的表达率较对照组低,差异有统计学意义(均P<0.05),但CD86、HLA-DR表达率2组差异无统计学意义(均P>0.05);NPC组CD83、CD1a、CD86和HLA-DR表达的平均荧光指数(MFI)均低于对照组,但差异无统计学意义(均P>0.05);对照组和NPC组外周血单核细胞均能诱导出DC,前者有较强的MLR诱导能力(P<0.05),而后者能力低下(P>0.05)。结论:NPC患者外周静脉血单核细胞来源的DC转化率较健康者低,且分化成熟能力也较低。
Objective: To observe the difference of morphology, phenotype and function of dendritic cells (DCs) derived from peripheral blood mononuclear cells in patients with nasopharyngeal carcinoma (NPC) and those with healthy DCs. Methods: DCs were isolated and cultured from peripheral blood of 12 NPC patients (NPC group) and 9 healthy controls (control group). The morphology of DCs was observed. The phenotypes of CD83, CD1a, CD86 and DCs were detected by flow cytometry HLA-DR, using 4, dimethylthiazole 2,5-diphenyltetrazolium bromide (MTT) colorimetric assay DC induced mixed lymphocyte reaction (MLR) capacity. Results: The morphology of DC in control group was more mature than that in NPC group. The expression rates of CD83 and CD1a in NPC group were lower than those in control group (all P <0.05), but there was no significant difference in the expression rates of CD86 and HLA-DR (All P> 0.05). The mean fluorescence index (MFI) of CD83, CD1a, CD86 and HLA-DR in NPC group were lower than those in control group, but the difference was not statistically significant DCs were induced by peripheral blood mononuclear cells in NPC group. The former had stronger ability of inducing MLR (P <0.05), while the latter had lower ability (P> 0.05). Conclusion: NPC patients with peripheral venous blood mononuclear cell-derived DC conversion rate is lower than healthy people, and the differentiation of mature ability is low.
