目的研究魟鱼骨素(RCE)对人乳腺癌细胞系MCF-7细胞生长周期的影响。方法以不同浓度RCE处理MCF-7细胞不同时间,采用MTT法检测各组MCF-7细胞增殖情况,流式细胞仪检测药物作用前后对细胞周期的变化,Western blot检测药物作用前后细胞周期素D1及p21蛋白表达。结果 RCE对MCF-7细胞增殖的抑制呈浓度依赖性和时间依赖性(P<0.05),且存在最大抑制浓度10μmol/L。随着RCE作用时间延长,S期细胞百分比明显增加,而G2/M期细胞比例明显减少(P<0.05),且细胞凋亡率随药物作用时间的延长明显升高(P<0.05)。随着药物作用时间延长,MCF-7细胞中的细胞周期素D1表达降低,p21蛋白表达逐渐增高(P<0.05)。结论 RCE抑制MCF-7细胞增殖,阻滞细胞从S期向G2期转化;其机制可能与调控细胞周期蛋白表达有关。RCE可能成为未来治疗乳腺癌药物之一。
Objective To investigate the effect of RCE on the growth of human breast cancer cell line MCF-7. Methods MCF-7 cells were treated with different concentrations of RCE for different time. The proliferation of MCF-7 cells was detected by MTT assay. The changes of cell cycle were detected by flow cytometry before and after drug treatment. The expression of cyclin D1 And p21 protein expression. Results The proliferation of MCF-7 cells was inhibited by RCE in a concentration-dependent and time-dependent manner (P <0.05), and the maximum inhibitory concentration was 10 μmol / L. With the prolongation of RCE, the percentage of cells in S phase increased significantly, while the proportion of cells in G2 / M phase decreased significantly (P <0.05), and the apoptosis rate increased with the prolongation of drug action time (P <0.05). With prolonged drug action, the expression of cyclin D1 and the expression of p21 in MCF-7 cells were increased (P <0.05). Conclusion RCE can inhibit the proliferation of MCF-7 cells and block the transformation of cells from S phase to G2 phase. The mechanism may be related to the regulation of the expression of cyclin. RCE may become one of the future treatment of breast cancer drugs.
