[目的]探讨在天然产物柠檬醛作用下,稻瘟病菌几丁质酶基因的表达量变化,以期揭示柠檬醛诱导稻瘟病菌的作用机制,为深入研究柠檬醛抗稻瘟病机理奠定基础。[方法]采用SYBR Green实时荧光定量PCR技术,定量检测不同浓度的柠檬醛处理后,稻瘟病菌菌丝中几丁质酶基因mRNA的表达情况。[结果]在柠檬醛诱导下,稻瘟病菌几丁质酶基因表达量显著升高,从时间效应和剂量效应来看,高质量浓度(200 mg/L)的柠檬醛在较短时间(6h)作用后,几丁质酶基因表达量升高最为显著,其上调幅度为对照组的52.5倍。[结论]表明稻瘟病菌几丁质酶基因可被柠檬醛诱导,且对柠檬醛非常敏感。由此推测,柠檬醛对稻瘟病菌的杀菌作用与几丁质酶密切相关。
[Objective] The research aimed to discuss the change of chitinase gene expression in Magnaporthe grisea with citral, a natural product, in order to reveal the mechanism of citral-induced Magnaporthe grisea and lay a foundation for further study on the mechanism of citral against blast. [Method] SYBR Green real-time fluorescence quantitative PCR was used to quantitatively detect the expression of chitinase gene mRNA in mycelia of Magnaporthe grisea after treated with different concentrations of citral. [Result] The chitinase gene expression of Magnaporthe grisea was significantly increased under citral induction. From the time effect and dose effect, citral with high concentration (200 mg / L) ), The expression of chitinase gene increased the most significant, the increase rate of 52.5 times the control group. [Conclusion] The chitinase gene of Magnaporthe grisea can be induced by citral and is very sensitive to citral. Therefore, it is speculated that the bactericidal effect of citral on Magnaporthe grisea is closely related to chitinase.
