目的 探讨 3 (4 5 ) 双甲基 2 噻唑 (2 5 ) 二苯基溴化四氮唑蓝 (MTT)法测定恶性肿瘤细胞对化疗药物的敏感性。方法 采用改良的MTT法检测了 189例恶性肿瘤标本对 9种化疗药物的体外敏感性。结果 5 1例胃癌细胞对化疗药物的敏感性 ,由高到低依次为丝裂霉素 (MMC) ,5 氟脲嘧啶 (5 Fu) ,顺铂 (DDP) ,阿霉素 (ADM ) ,阿糖胞苷 (Ara C) ,平阳霉素 (PYM ) ,甲氨喋呤 (MTX) ,足叶乙甙 (VP 16 ) ,长春新碱 (VCR ) ;75例结直肠癌细胞对化疗药物的敏感性 ,由高到低依次为 5 Fu ,MMC ,PYM ,VP 16 ,MTX ,VCR ,DDP ,Ara C ,ADM。43例肝癌细胞对化疗药物的敏感性 ,由高到低依次为DDP ,MMC ,5 Fu ,ADM ,MTX ,Ara C ,VP 16 ,PYM ,VCR ;2 0例乳癌细胞对化疗药物的敏感性 ,由高到低依次为 5 Fu ,MMC ,DDP ,ADM ,MTX ,PYM ,VCR ,Ara C ,VP 16。结论 MTT法快速、简便、敏感性高、可评价率高 ,适用于恶性肿瘤的药敏试验 ,对临床用药有指导意义
Objective To investigate the sensitivity of 3 (45) bismethyl-2-thiazole (25) diphenyltetrazolium bromide (MTT) method to determine the sensitivity of malignant cells to chemotherapy drugs. Methods The in vitro sensitivity of 189 malignant tumor specimens to 9 chemotherapeutic agents was tested using a modified MTT assay. Results The sensitivities of 51 gastric cancer cells to chemotherapeutic drugs were mitomycin (MMC), 5 fluorouracil (5 Fu), cisplatin (DDP), and adriamycin (ADM) from high to low. Cytosine (Ara C), Pingyangmycin (PYM), Methotrexate (MTX), Etoposide (VP 16), Vincristine (VCR); 75 cases of colorectal cancer cells sensitive to chemotherapy drugs Sex, from high to low, followed by 5 Fu, MMC, PYM, VP 16, MTX, VCR, DDP, Ara C, ADM. The sensitivity of 43 cases of hepatocellular carcinoma cells to chemotherapeutic drugs, from high to low, were DDP, MMC, 5 Fu, ADM, MTX, Ara C, VP 16, PYM, and VCR; the sensitivity of 20 breast cancer cells to chemotherapy drugs. The order from high to low is 5 Fu, MMC, DDP, ADM, MTX, PYM, VCR, Ara C, VP 16. Conclusion The MTT method is rapid, simple, sensitive, and highly evaluable. It is suitable for drug susceptibility testing of malignant tumors and has guiding significance for clinical drug use.
