采用复乳溶剂扩散-挥发法制备牛血清白蛋白的聚乳酸纳米粒,分别用自制的水溶性壳聚糖和聚乙烯醇进行表面修饰,并考察了两种纳米粒的粒径、电位、表面亲水性和在小鼠血液中的存留量-时间变化情况。结果表明,制备的两种表面修饰纳米粒粒径均为100~200 nm。用水溶性壳聚糖修饰能有效提高纳米粒的表面亲水性,并调节表面电荷至接近中性(5.2 mV)。用DAS 2.0软件计算得水溶性壳聚糖修饰纳米粒的血液半衰期为6.9 h,明显高于聚乙烯醇修饰纳米粒(0.3 h)。
The polylactic acid nanoparticles with bovine serum albumin (BSA) were prepared by the complex emulsion solvent diffusion-volatilization method. The surfaces were modified with homemade water-soluble chitosan and polyvinyl alcohol. The particle size, potential, surface Hydrophilicity and retention in mouse blood-time changes. The results show that the prepared two kinds of surface-modified nanoparticles have a particle size of 100-200 nm. Water-soluble chitosan modification can effectively improve the surface hydrophilicity of nanoparticles and adjust the surface charge to near neutral (5.2 mV). The half-life of water-soluble chitosan-modified nanoparticles was 6.9 h calculated by DAS 2.0 software, which was significantly higher than that of polyvinyl alcohol-modified nanoparticles (0.3 h).
