目的:观察姜黄素对APPswe/PS1dE9双转基因小鼠Aβ42及其降解酶NEP表达的影响。方法:将3月龄的APP-swe/PS1dE9双转基因小鼠随机分为模型组、阳性对照组、姜黄素高、中、低剂量组。灌胃3个月后,应用Morris水迷宫、免疫组织化学、Western blot等方法进行检测。结果:行为学检测,治疗组与模型组相比定位航行实验和空间探索实验存在不同程度的差异(P<0.01或P<0.05)。Aβ42及其降解酶NEP表达,模型组小鼠大脑海马CA1区较正常对照组Aβ42阳性细胞明显增加(P<0.01)。结论:姜黄素能通过增加Aβ42降解酶NEP表达、降低Aβ42蛋白,进而改善APPswe/PS1dE9双转基因小鼠的学习记忆能力。
Objective: To observe the effect of curcumin on the expression of Aβ42 and NEP in APPswe / PS1dE9 double transgenic mice. Methods: Three-month-old APP-swe / PS1dE9 double transgenic mice were randomly divided into model group, positive control group, curcumin high, medium and low dose groups. Three months after gavage, Morris water maze, immunohistochemistry, Western blot and other methods were used for detection. Results: Compared with the model group, the navigation test and the space exploration experiment in the treatment group showed different degrees of difference (P <0.01 or P <0.05). The expression of Aβ42 and its degrading enzyme NEP in hippocampal CA1 region of hippocampal CA1 region in model group was significantly higher than that in normal control group (P <0.01). Conclusion: Curcumin can improve the learning and memory abilities of APPswe / PS1dE9 double transgenic mice by increasing the NEP expression of Aβ42 degrading enzyme and decreasing Aβ42 protein.
