目的研究膀胱癌细胞系BIU-87体外转染pCH510后,CH50多肽的表达及其对卡介苗(bacilluscalmette-guerin,BCG)抑瘤作用的影响。方法在LipofectimineTM2000的介导下,将质粒pCH510体外转染给BIU-87细胞,采用免疫组织化学S-P法和WesternBlot法鉴定CH50多肽的表达;通过四唑盐比色法(MTT法)观察pCH510转染BIU-87对BCG细胞毒作用的影响。结果BIU-87细胞转染pCH510后阳性表达CH50多肽,对照组则不表达;CH50多肽阳性表达增强了BCG对BIU-87的细胞毒性作用(P<0.05)。结论转染pCH510后,BIU-87细胞表达CH50多肽,并因此促进了BCG对膀胱癌细胞的抑制作用。
Objective To study the expression of CH50 polypeptide after transfecting pCH510 in bladder cancer cell line BIU-87 in vitro and its effect on the inhibition of bacillus Calmette-guerin (BCG). Methods The plasmid pCH510 was transfected into BIU-87 cells with LipofectimineTM 2000. The expression of CH50 was detected by immunohistochemical SP method and Western Blot. The expression of CHCH1 was detected by MTT method. Effect of BIU-87 on BCG cytotoxicity. Results The expression of CH50 peptide in BIU-87 cells transfected with pCH510 was not detected in the control group. The positive expression of CH50 peptide enhanced the cytotoxicity of BCG to BIU-87 cells (P <0.05). Conclusion After transfection with pCH510, BIU-87 cells express CH50 polypeptide and thus promote the inhibitory effect of BCG on bladder cancer cells.
