目的:尝试以电视辅助胸腔镜外科技术(VATS)为新方法,探索建立慢性微创心肌梗死模型的制作方法。方法:在电视辅助胸腔镜下找到家犬的冠状动脉前降支后,以钛夹钳夹,造成心肌梗死并进行长期观察,系统监测了模型组与对照组的肢体导联ST段抬高值、全血白细胞计数(WBC)、血清OMP、肌酸激酶(CK)、心肌肌钙蛋白I(cTnI)含量、梗塞区面积占心室面积的百分比(%)等一系列生理、生化指标,同时观察了两组的Masson三色染色病理学表现。结果:模型家犬的ST段抬高值、WBC、血清OMP、CK、cTnI含量、梗塞区面积占心室面积的百分比(%)与对照组比较,均有显著性差异。Masson三色染色与cTnI免疫组化染色也为梗死区的形成提供了直接的证据。结论:VATS下钛夹夹闭犬冠脉前降支能够建立家犬的慢性微创心肌梗死模型。
OBJECTIVE: To explore a method of making a model of chronic minimally invasive myocardial infarction by using video assisted thoracoscopic surgery (VATS) as a new method. Methods: After the anterior descending coronary artery was found in the video-assisted thoracoscope, the myocardial infarction was observed with titanium clamp and the long-term observation was performed. The ST segment elevation of the limb lead in the model group and the control group was monitored (WBC), serum OMP, creatine kinase (CK), cardiac troponin I (cTnI), the percentage of infarct area to the ventricular area (%) and a series of physiological and biochemical indicators were observed Masson’s trichrome staining pathology was performed in both groups. Results: The ST segment elevation, WBC, serum OMP, CK, cTnI, percentage of infarction area to ventricular area (%) in model dogs were significantly different from those in control group. Masson trichrome staining and cTnI immunohistochemical staining also provide direct evidence of infarct formation. CONCLUSION: Dogs under VATS can establish a chronic minimally invasive myocardial infarction model by occluding canine coronary artery in dogs.
