三氧化二砷是治疗急性早幼粒细胞白血病的特效药。三价砷是巯基(-SH)的络合剂,砷化物与-SH结合后可能通过多条途径诱导细胞凋亡,此外,砷与-SH结合还直接导致细胞内的还原性谷胱甘肽耗竭,使活性氧产生增多。研究发现,还原性谷胱甘肽(GSH)水平与三氧化二砷对急性早幼粒细胞白血病、多发性骨髓瘤、淋巴瘤等细胞株的疗效呈负相关,降低细胞内GSH水平会明显增强三氧化二砷的疗效。
