目的:研究中老年人骨髓间质干细胞(MSCs)的生物学特性。方法:用Ficoll-Paque分离不 同年龄段供者MSCs体外培养,进行诱导分化,用流式细胞术检测表面标志,核型分析观察细胞染 色体的稳定性,裸鼠实验、软琼脂实验观察致瘤性。结果:中老年人每毫升骨髓分离的单个核细 胞量和MSCs增殖速度与青年人无显著差异;中老年人不同代次的MSCs诱导后具有向成骨细胞 和成脂细胞分化的能力;分离培养的MSCs表达CD44,不表达CD34;传至第10代核型未见异常, 未见致瘤性。结论:中老年人MSCs有较强的体外增殖能力和多向分化潜能,遗传背景稳定。
Objective: To study the biological characteristics of bone marrow mesenchymal stem cells (MSCs) in the elderly. Methods: MSCs from different age groups were isolated and cultured in vitro with Ficoll-Paque for differentiation. The surface markers were detected by flow cytometry. The karyotype analysis was used to observe the stability of the cells. Nude mice experiments and soft agar experiments were used to observe the tumorigenicity . Results: There was no significant difference in the number of mononuclear cells isolated from bone marrow per milliliter and the proliferation rate of MSCs in middle-aged and elderly people. MSCs of different generations in middle-aged and old people had the ability to differentiate into osteoblasts and adipogenic cells; Of MSCs express CD44, but do not express CD34; there is no abnormality in the passage to the 10th generation, no tumorigenicity. Conclusion: MSCs in middle-aged and elderly people have strong ability of proliferation and multi-directional differentiation in vitro, and their genetic background is stable.
