目的初步探讨不同剂量双酚A对SD大鼠卵巢的毒性作用。方法实验将48只4周龄SD雌性大鼠随机分四组,分别予以不同剂量BPA灌胃染毒,隔日一次,染毒180 d。用酶联免疫法测定雌二醇(E2)、孕酮(P)、睾丸酮(T)3种主要激素水平值。取大鼠卵巢组织固定切片,做HE染色;用免疫组化方法检测雌激素受体ER和P53在大鼠卵巢中的分布及表达。结果与对照组比较,各染毒组大鼠血清中雌二醇(E2)和孕酮(P)水平值增高,睾丸酮(T)水平值降低,并随染毒剂量的增高而降低,呈负相关。病理组织切片显示卵泡颗粒层排列有明显的改变;免疫组化结果表明:ER和P53在SD大鼠卵巢细胞中的表达增强,呈现出剂量-效应关系。结论 BPA对SD大鼠具有内分泌干扰作用,可改变SD大鼠血清中雌二醇(E2)、孕酮(P)、睾酮(T)的正常水平值,增强卵巢中ER和P53的表达,影响SD大鼠卵巢细胞的正常增殖。
Objective To investigate the toxic effects of different doses of bisphenol A on the ovaries of SD rats. Methods Forty-eight 4-week-old female Sprague-Dawley rats were randomly divided into four groups and given different doses of BPA intragastrically. The rats were treated once daily for 180 days. The levels of three major hormones, estradiol (E2), progesterone (P) and testosterone (T), were determined by enzyme-linked immunosorbent assay. The fixed ovarian tissue sections were taken and HE staining was performed. The distribution and expression of estrogen receptor ER and P53 in rat ovary were detected by immunohistochemistry. Results Compared with the control group, the levels of estradiol (E2) and progesterone (P) in serum increased and the levels of testosterone (T) decreased in all the treated groups, and decreased with the increase of the dosage Related. Histological sections showed that the arrangement of follicular granulosa layers changed significantly. The results of immunohistochemistry showed that the expression of ER and P53 in ovary cells of SD rats was increased, showing a dose-response relationship. Conclusion BPA has an endocrine disrupting effect on SD rats, which can change the normal serum estradiol (E2), progesterone (P) and testosterone (T) levels in SD rats and enhance the expression of ER and P53 in ovary Normal proliferation of SD rat ovarian cells.
