目的:采用核磁共振(MRI)动态观测白藜芦醇预处理对大鼠局部脑缺血4h和24h时脑水肿和梗死脑组织体积的影响。方法:20只SD大鼠随机分为白藜芦醇预处理组(简称预处理组,n=10)和对照组(n=10)。预处理组手术前10天,每日按照30 mg/kg腹腔注射白藜芦醇溶液。对照组给予同等体积的白藜芦醇溶剂注射10天。10日后行左侧大脑中动脉永久缺血术(MCAO)。术后4 h和24 h分别采用MRI对大鼠纹状体区和顶颞叶区进行T1-SE、T2-TSE、T2-map-SE以及DW-EPI序列的扫描。采用ImageJ软件分析扫描图像中4个感兴趣区域(左侧纹状体、顶颞叶,右侧纹状体、顶颞叶)的表观弥散系数(ADC)值、T2横向弛豫时间(T2-relaxation time,T2RT)值的变化。术后24 h处死大鼠,TTC(2,3,5-氯化三苯基四氮唑)染色脑片,UTHSCSA Image Tool软件计算梗死脑组织体积的比例。结果:①预处理组大鼠缺血4 h时纹状体和顶颞叶相对ADC值较对照组显著增加(P<0.01),左侧T2RT值显著减小(P<0.05)。②预处理组大鼠缺血24 h时纹状体和顶颞叶相对ADC值较对照组均显著增加(P<0.01),左侧T2RT值也显著减小(P<0.05)。③预处理组大鼠梗死组织脑体积比例为0.313±0.047,对照组为0.396±0.021。两组相比有显著性差异(P<0.01)。结论:白藜芦醇预处理对大鼠脑缺血后4 h和24 h时均有减轻脑水肿的作用,并且可以有效减少脑梗死体积,从而起到神经保护作用。
Objective: To observe the effects of resveratrol pretreatment on brain edema and infarct volume at 4h and 24h after focal cerebral ischemia in rats by using MRI. Methods: Twenty SD rats were randomly divided into resveratrol pretreatment group (n = 10 for short) and control group (n = 10). Pretreatment group 10 days before surgery, daily dose of 30 mg / kg resveratrol solution. The control group was given the same volume of resveratrol solvent injection for 10 days. 10 days after the left middle cerebral artery permanent ischemia (MCAO). The images of T1-SE, T2-TSE, T2-map-SE and DW-EPI were scanned by MRI at 4 h and 24 h after operation. ImageJ software was used to analyze the apparent diffusion coefficient (ADC) values of four regions of interest (left striatum, apex of the temporal lobe, right striatum, apex of the temporal lobe), T2 transverse relaxation time (T2 -relaxation time, T2RT) value changes. Twenty-four hours after operation, TTC (2,3,5-triphenyltetrazolium chloride) stained brain slices were taken and the proportion of infarcted brain tissue volume was calculated by UTHSCSA Image Tool software. Results: (1) Compared with the control group, the relative ADC value of striatum and the top temporal lobe increased significantly (P <0.01) and the left T2RT decreased significantly (P <0.05) at 4 h after ischemia in the preconditioning group. ② Compared with the control group, the ADC value of striatum and the top temporal lobe in ischemic group at 24h after preconditioning was significantly increased (P <0.01), and the left T2RT decreased significantly (P <0.05). ③ The pretreatment group infarct brain volume ratio was 0.313 ± 0.047, the control group was 0.396 ± 0.021. There was significant difference between the two groups (P <0.01). CONCLUSION: Resveratrol preconditioning can relieve cerebral edema at 4 h and 24 h after cerebral ischemia in rats, and can effectively reduce the volume of cerebral infarction and thus play a neuroprotective role.
