目的观察重症急性胰腺炎大鼠白细胞粘附变化以及胰腺细胞间粘附分子-1(intracel-lular adhesion molecules,ICAM-1)的表达变化。方法 SD 雄性大鼠36只,随机分为假手术组(SO 组)和重症急性胰腺炎组(SAP 组),每组18只。观察胰腺白细胞粘附数目,取腹主动脉血测血清淀粉酶,用 SYBR-Green 荧光定量 PCR 的方法检测胰腺组织 ICAM-1的 mRNA 表达,取胰腺组织行病理组织学观察。结果 SAP 组与 SO 组相比,各时间点血清淀粉酶,白细胞粘附数目、ICAM-1表达水平明显增高,P<0.05,胰腺坏死程度随着观察时间点的延长而加重,12 h 坏死程度最重,而白细胞粘附数目、ICAM-1表达在6 h 最高。结论白细胞粘附与重症急性胰腺炎的发生发展有一定的关系,ICAM-1参与白细胞粘附。
Objective To observe the changes of leukocyte adhesion and the expression of intercellular adhesion molecule-1 (ICAM-1) in severe acute pancreatitis in rats. Methods Thirty-six SD male rats were randomly divided into sham operation group (SO group) and severe acute pancreatitis group (SAP group), with 18 rats in each group. The number of pancreatic leukocyte adhesion was observed. The serum amylase was measured by abdominal aorta. The expression of ICAM-1 mRNA in pancreatic tissue was detected by SYBR-Green fluorescence quantitative PCR. Pathological histology was used to observe the pancreatic tissue. Results Compared with SO group, the number of amylase, leukocyte adhesion and the expression of ICAM-1 in SAP group were significantly increased at each time point (P <0.05), the degree of pancreatic necrosis increased with the prolongation of observation time, and the degree of necrosis 12 h The heaviest, while the number of leukocyte adhesion, ICAM-1 expression at 6 h highest. Conclusion Leukocyte adhesion has a certain relationship with the occurrence and development of severe acute pancreatitis. ICAM-1 is involved in leukocyte adhesion.
