目的 研究核因子-κB(NF-κB)在中药单体黄芩甙体外诱导骨髓基质细胞分化为神经细胞的作用。方法 实验分为黄芩甙组和对照组。黄芩甙组采用黄芩甙作为主要诱导剂,诱导成年大鼠骨髓基质细胞6d。对照组仅用无血清培养基诱导。采用间接免疫荧光染色检测神经细胞标记蛋白和NF-κB亚基P65的表达。结果黄芩甙诱导6d,骨髓基质细胞形成较典型的神经细胞形态,同时表达多种神经细胞标记蛋白;对照组未见神经细胞标记蛋白表达。而且,对照组诱导6d,P65信号出现核移位,胞浆中信号明显减弱到(18.4±3.0)%,黄芩甙组P65信号(84.8±3.0)%主要仍在胞浆。结论 黄芩甙可以抑制NF-κB的活化,在骨髓基质细胞诱导分化为神经细胞过程中可能起到一定的作用。 [中国当代儿科杂志,2003,5(1):1—4]
Objective To investigate the role of nuclear factor-κB (NF-κB) in inducing the differentiation of bone marrow stromal cells into nerve cells in vitro. Methods The experiment was divided into astragalus group and control group. The Huangqi group used astragalus as the main inducer to induce bone marrow stromal cells in adult rats for 6 days. The control group was induced only with serum-free medium. Indirect immunofluorescence staining was used to detect the expression of neuronal marker protein and NF-κB subunit P65. Results After induction of Astragalus membranaceus for 6 days, bone marrow stromal cells formed more typical neuronal cell morphology and expressed multiple neuronal cell marker proteins. No neuronal marker protein expression was observed in the control group. Furthermore, the nuclear translocation of P65 signal was observed in the control group at 6 days after induction, and the signal in the cytoplasm was significantly reduced to (18.4±3.0)%. The P65 signal in the Astragalus group was (84.8±3.0)% mainly in the cytoplasm. Conclusion Astragalus can inhibit the activation of NF-κB and may play a role in the induction of differentiation of bone marrow stromal cells into neural cells.
