目的:观察卷柏醇提物所得各部位对糖尿病大鼠胰岛组织形态及骨骼肌葡萄糖转运蛋白-4(GLUT-4)表达的影响。方法:通过腹腔注射链脲佐菌素(STZ,65mg/kg)诱导糖尿病大鼠模型,观察卷柏各部位给药7d后,空腹血糖、餐后4h血糖变化,观察胰岛组织HE染色切片,并通过免疫组化的方法观察GLUT-4的表达情况。结果:卷柏80%乙醇部位组大鼠空腹血糖及餐后4h血糖与模型组比较均显著降低(P<0.05),GLUT-4的表达量比模型组显著增加(P<0.01),胰岛组织形态与模型组比较也有所改善。结论:卷柏80%乙醇部位可能是卷柏降糖的活性部位,其降糖作用机制可能与修复受损的胰岛组织,增强骨骼肌GLUT-4表达,进而增强机体胰岛素敏感性有关。
OBJECTIVE: To observe the effect of ethanol extract from Betula platyphylla on the islet morphology and the expression of GLUT-4 in diabetic rats. Methods: Diabetic rats were induced by intraperitoneal injection of streptozotocin (STZ, 65mg / kg), and the fasting blood glucose and postprandial blood glucose were observed at 7 days after administration. The expression of GLUT-4 was observed by immunohistochemistry. Results: Compared with the model group, the fasting blood glucose and the 4h postprandial blood glucose in the tissue of Selaginel group were significantly decreased (P <0.05), the expression of GLUT-4 was significantly increased (P <0.01) Form and model groups also improved. CONCLUSION: 80% ethanolic cytoplasm may be the active site of Betula platyphylla hypoglycemic. The hypoglycemic mechanism may be related to the repair of impaired islet tissue, the enhancement of GLUT-4 expression in skeletal muscle, and the enhancement of insulin sensitivity.
