目的:评价阿霉素和rHuTNF抗人结肠癌细胞株的序贯作用。方法:采用克隆生成试验(Hamburger-Salmon法),研究DOX和rHuTNF序贯作用于COLO205细胞株后的抗增殖效果。结果:对结肠癌细胞系COLO205,存在DOX-rHuTNF序贯抗增殖作用;多数联合给药情况下出现相加/协同作用。结论:DOX存在时,TNF的抗瘤作用增强,从而支持DOX可干扰细胞保护机制以使瘤细胞对TNF细胞毒作用更为敏感的理论。
Objective: To evaluate the sequential effect of doxorubicin and rHuTNF against human colon cancer cell lines. Methods: The antiproliferative effect of sequential DOX and rHuTNF treatment on COLO205 cell line was investigated by the method of Hamburger-Salmon assay. RESULTS: There was DOX-rHuTNF sequential antiproliferative effect on colon cancer cell line COLO205; the additive / synergistic effects were observed in the majority of combination therapy. CONCLUSION: The antitumor effect of TNF is enhanced in the presence of DOX, thus supporting the theory that DOX may interfere with cytoprotective mechanisms to make tumor cells more sensitive to TNF cytotoxicity.
