目的:本文以养殖河豚鱼肝脏为实验材料,对其鱼油的提取工艺和EPA、DHA的检测进行了系统的研究。方法:采用超声提取法对鱼油的提取方法进行研究,设计L9(34)正交表,采用毛细管气相色谱法检测,以鱼油提取率评价指标,优选最佳提取条件。结果:超声时间、正己烷用量、提取次数对鱼油提取率均有影响,其中超声时间和溶剂用量有显著影响,最佳提取工艺为用河豚肝脏4倍量的正己烷,超声提取2次,每次30m in。超声提取法优于传统法,且鱼油的理化指标均符合标准。结论:按照最优处方采用超声提取法提取鱼油是可行的,且用超声法提取的鱼油中EPA和DHA的含量最高。
Objective: This article systematically studied the extraction process of fish oil and the detection of EPA and DHA from the cultured puffer fish liver. Methods: Ultrasonic extraction method was used to study the extraction method of fish oil. L9 (34) orthogonal table was designed. Capillary gas chromatography was used to determine the extraction index of fish oil. The best extraction conditions were optimized. RESULTS: Ultrasonic time, n-hexane dosage and extraction times all had effects on the extraction rate of fish oil. Ultrasonic time and solvent dosage had a significant effect. The best extraction process was using bufo liver 4 times the amount of n-hexane, ultrasonic extraction twice, each 30m in. The ultrasonic extraction method is superior to the traditional method, and the physical and chemical indicators of fish oil are in line with the standard. Conclusion: It is feasible to use ultrasonic extraction method to extract fish oil according to the best prescription. The content of EPA and DHA in fish oil extracted by ultrasonic method is the highest.
