目的:观察兔骨髓基质细胞(MSC)的体外生长特性,为用于骨/软骨组织工程的功能细胞提供实验基础.方法:于胫骨近端抽取骨髓,分离MSC,原代培养,形成CFU-F,以1×10~4个细胞/cm~2传代培养5代,绘制细胞生长曲线,计算倍增时间,测定分裂指数和贴壁率.结果:第1~5代传代细胞生长曲线基本相同,第3代细胞倍增时间为24h,分裂指数最高达19%,12h内95%细胞贴壁.结论:在本实验培养条件下,MSC体外生长稳定,增殖速度较快,接种成活率较高,适应性强,可用于自体骨/软骨组织工程.
OBJECTIVE: To observe the in vitro growth characteristics of rabbit bone marrow stromal cells (MSC) and provide experimental basis for the functional cells used in bone / cartilage tissue engineering.Methods: Bone marrow was harvested from the proximal tibia and MSCs were isolated and primary cultured to form CFU-F , The cells were subcultured for 5 passages at 1 × 10 ~ 4 cells / cm ~ 2, the cell growth curve was plotted, the doubling time was calculated, the index of division and the rate of attachment were measured.Results: The growth curves of passage 1 ~ 5 were basically the same, The third generation of cells doubling time was 24h, the dividing index was up to 19%, 95% cells within 12h adherent.Conclusion: Under the experimental culture conditions, MSC growth in vitro stable, rapid proliferation, vaccination survival rate, adaptability Strong, can be used for autologous bone / cartilage tissue engineering.
