目的观察来氟米特(LEF)对子宫内膜异位症(EMT)模型大鼠异位内膜及白细胞介素-1β(IL-1β)的影响,探讨LEF对EMT的作用机制。方法自体移植法建立EMT大鼠模型,观察LEF对大鼠异位子宫内膜形态学的影响;免疫组化方法检测异位子宫内膜IL-1β表达;放免法检测腹腔液IL-1β含量。结果 LEF组大鼠的异位子宫内膜体积明显缩小(P<0.01),异位子宫内膜IL-1β的表达也明显低于对照组(P<0.05),腹腔液中IL-1β水平也较对照组和空白组明显降低(P<0.05)。结论 LEF可抑制EMT模型大鼠异位病灶的生长,其作用机制可能是通过减少IL-1β等细胞因子的生成而实现的。
Objective To observe the effect of leflunomide (LEF) on ectopic endometrium and interleukin-1β (IL-1β) in rats with endometriosis (EMT) and to explore the mechanism of LEF on EMT. Methods EMT rat model was established by autograft. The morphology of ectopic endometrium in rats was observed by LEF. The expression of IL-1β in ectopic endometrium was detected by immunohistochemistry. The content of IL-1β in peritoneal fluid was detected by radioimmunoassay. Results The volume of ectopic endometrium in LEF group was significantly reduced (P <0.01), and the expression of IL-1β in ectopic endometrium was significantly lower than that in control group (P <0.05). The level of IL-1β in peritoneal fluid Compared with the control group and the blank group was significantly lower (P <0.05). Conclusion LEF can inhibit the growth of ectopic lesions in EMT model rats, and its mechanism may be through the reduction of the production of cytokines such as IL-1β.
