目的探讨过氧化物酶体增殖因子激活受体γ(PPAR-γ)、环氧合酶-2(COX-2)在子宫内膜癌中的表达及其超微结构定位。方法免疫组织化学法检测正常子宫内膜、子宫内膜增生症、子宫内膜癌组织中PPAR-γ、COX-2的表达;胶体金免疫电镜技术观察COX-2在子宫内膜癌细胞中的超微结构定位。结果免疫组织化学结果显示,1.PPAR-γ在正常子宫内膜、子宫内膜增生症中不表达,在子宫内膜癌中表达;2.COX-2在正常子宫内膜、子宫内膜增生症、子宫内膜癌3种组织中均表达,但表达不同,3组间差异有显著性(P<0.01);3.子宫内膜癌组织中PPAR-γ的表达与COX-2的表达显著相关(P<0.05)。免疫电镜结果显示,子宫内膜癌细胞的内质网和核膜上有黑色圆形集中的胶体金颗粒分布。结论PPAR-γ和COX-2可能参与子宫内膜癌的发生、发展。
Objective To investigate the expression of peroxisome proliferator - activated receptor γ (PPAR - γ) and cyclooxygenase - 2 (COX - 2) in endometrial carcinoma and their ultrastructural localization. Methods Immunohistochemistry was used to detect the expression of PPAR-γ and COX-2 in normal endometrium, endometrial hyperplasia and endometrial carcinoma. The expression of COX-2 and COX-2 in endometrial carcinoma were detected by colloidal gold immunoelectron microscopy Ultrastructural positioning. Results The results of immunohistochemistry showed that 1.PPAR-γ was not expressed in normal endometrium and endometrial hyperplasia and was expressed in endometrial carcinoma.2.COX-2 in normal endometrium and endometrial hyperplasia (P <0.01). 3. The expression of PPAR-γ and COX-2 in endometrial carcinoma were significantly higher than that in endometrial carcinoma Related (P <0.05). Immunoelectron microscopy showed that the endoplasmic reticulum and the nuclear membrane of endometrial cancer cells had a concentrated distribution of colloidal gold particles with a round black circle. Conclusion PPAR-γ and COX-2 may participate in the occurrence and development of endometrial cancer.
