目的研究硫酸高乌甲素对人宫颈癌Hela细胞增殖、周期和凋亡的影响及其可能作用机制。方法采用细胞计数试剂盒8(CCK-8)法考察硫酸高乌甲素对Hela细胞的增殖抑制作用,并计算其半抑制浓度(IC_(50));运用流式细胞技术,采用碘化丙啶(PI)单标记法和Annexin V-FITC/PI双标记法检测硫酸高乌甲素作用后Hela细胞的细胞周期及细胞凋亡情况。结果硫酸高乌甲素对人宫颈癌Hela细胞的增殖具有抑制作用,且这种抑制作用具有明显的量效关系(r=0.990 5,P<0.01),其对Hela细胞的IC_(50)为0.421 mg·mL~(-1)。经硫酸高乌甲素作用后,Hela细胞出现了明显的G_0/G_1期阻滞,且细胞凋亡率随硫酸高乌甲素浓度的增大而逐渐升高。结论硫酸高乌甲素能抑制Hela细胞的生长,可改变细胞周期分布并诱导其凋亡。
Objective To study the effect of lappaconitine on proliferation, cycle and apoptosis of human cervical cancer Hela cells and its possible mechanism. Methods The proliferation inhibition effect of Lappaconitine on Hela cells was investigated by CCK-8 method and the half-inhibitory concentration (IC 50) was calculated. Using flow cytometry, (PI) single labeling method and Annexin V-FITC / PI double labeling method were used to detect the cell cycle and apoptosis of Hela cells. Results Lappaconitine had an inhibitory effect on the proliferation of human cervical cancer Hela cells, and the inhibitory effect was significant (r = 0.990 5, P <0.01). The IC 50 of Hela cells was 0.421 mg · mL -1. After lappaconitine sulfate treatment, Hela cells appeared obvious G_0 / G_1 phase arrest, and the apoptosis rate increased gradually with the increase of lappaconitine concentration. Conclusion Lappaconitine can inhibit the growth of Hela cells, change the cell cycle distribution and induce apoptosis.
