目的 :探讨黄芪对过敏性豚鼠哮喘模型中核因子 - κB(nuclear factor- kappa B,NF- κB)表达的影响。方法 :将 30只豚鼠随机分为 3组 (正常组、哮喘组、治疗组 ) ,每 10只。用 10 %卵清蛋白 (OVA)溶液 1ml腹腔注射致敏 ,2周后哮喘组及治疗组用 1% OVA溶液超声雾化吸入致其哮喘发作 ,1次 / d,共 2周 ;正常组则喷入生理盐水。治疗组每次诱喘后给予腹腔注射黄芪注射液 5 g/ kg,1次 / d,共 2周。然后分别测定 3组肺功能并观察气道壁嗜酸性粒细胞 (EOS)浸润情况 ;用免疫组织化学染色方法观察 NF- κB在豚鼠肺组织中的表达变化。结果 :NF- к B主要表达在气道壁内的细胞中 ,3组胞核阳性的细胞数占气道上皮细胞数百分比分别为 (14 .3± 2 .6 ) %、(32 .5± 5 .8) %和 (2 0 .4± 3.3) %。哮喘组 NF- к B胞核阳性的细胞比例显著高于正常组 (P <0 .0 1) ,治疗组 NF- к B胞核阳性的细胞比例显著低于哮喘组 (P <0 .0 1)。结论 :黄芪能显著抑制过敏性哮喘豚鼠气道壁内细胞 NF- κB的表达 ,提示黄芪抑制 NF- κB的表达可能是黄芪治疗哮喘的作用机制之一
Objective: To investigate the effect of astragalus on the expression of nuclear factor-kappa B (NF-κB) in asthmatic model of allergic guinea pigs. Methods: 30 guinea pigs were randomly divided into 3 groups (normal group, asthma group, treatment group), each 10 rats. The mice were sensitized by intraperitoneal injection of 1ml ovalbumin (OVA) solution (10ml). After 2 weeks, the asthma group and the treatment group were challenged with 1% OVA solution by ultrasonic atomization for 1 week for 2 weeks. In the normal group Spray saline. Each treatment group received intraperitoneal injection of astragalus injection 5 g / kg, once a day for 2 weeks. Then the lung function was measured in 3 groups and the infiltration of eosinophils in the airway wall was observed. The expression of NF-κB in the lung tissue of guinea pigs was observed by immunohistochemical staining. Results: NF-к B was mainly expressed in the airway wall cells. The numbers of positive cells in the three groups were (14.3 ± 2.66)%, (32.5 ± 5 .8)% and (20.4 ± 3.3)%. The proportion of NF-к B-positive cells in the asthmatic group was significantly higher than that in the normal group (P <0.01). The proportion of NF-к B-positive cells in the asthmatic group was significantly lower than that in the asthmatic group (P <0.01) ). Conclusion: Astragalus can significantly inhibit the expression of NF-κB in the airway wall cells of asthmatic guinea pigs, suggesting that the inhibition of NF-κB expression may be one of the mechanisms of astragalus in the treatment of asthma
